What are the practical differences between Main Board and Main Board (Short)? Do they require different preparation?

The key difference is that the Main Board selects Regular Officers and the Short selects Reserve or Professionally qualified officers.

In terms of preparation, I would prepare for both in the same way. A lot of the testing is looking at your innate abilities and personality but you can control certain elements such as fitness and current affairs knowledge. I would also practice mental arithmetic as this makes some of the planning exercises easier. There is a lot of public speaking as part of the assessment (and is a large part of being an officer anyway!) so it might be worth taking the opportunity to practice speaking in front of people if opportunities present themselves.

Ultimately they are not looking for the finished product so just be confident and be yourself!
